Palos de la Frontera is the most important stop on the Ruta de los Lugares Colombinos. On 3 August 1492, the expedition led by Christopher Columbus and the Pinzón brothers set out from the Puerto Histórico de Palos.
In the square of the Iglesia Parroquial de San Jorge Mártir, the Royal Pragmatic Ordinance was read out in May 1492. It required Palos to hand over two caravels to Christopher Columbus and recruit local sailors.
The Iglesia Parroquial de San Jorge Mártir is a Gothic-Mudéjar building with three Mudéjar naves and a Gothic apse. It was declared a national monument in 1931.
The Casa natal, Museo Martín Alonso Pinzón was built in the mid-15th century. It was the birthplace of Martín Alonso, Vicente Yáñez and Francisco Martín Pinzón. The house is currently used for the research, preservation and permanent exhibition of documents, bibliography and objects relating to the history of the Pinzón family and Palos de la Frontera in connection with the discovery of America.
La Fontanilla is located on an ancient Roman well ring. It is protected by a small Mudéjar temple, or tetrapylon, built of bricks in the 13th century.
The pottery workshop discovered at the Puerto Histórico de Palos had twelve kilns. It is considered the most complete and best-preserved pottery workshop in Europe from the 15th and 16th centuries.

The Paraje Natural Lagunas de Palos y Las Madres nature area covers 693 hectares. It is located in the municipalities of Palos de la Frontera and Moguer in the province of Huelva.
The José Celestino Mutis Botanical Park covers 12 hectares and is crossed by footpaths leading to a perimeter route and an inner route. The park is dedicated to presenting Spanish and Ibero-American flora.
